Ninhydrin reagent is said to be stable for about 2 weeks. However, since its color development decreases gradually with time, a certain amount of an amino acid other than the constituent amino acids in the analyte protein should be added as an internal standard, and the analytical value should be corrected using the recovery rate of the internal standard. The detection limit is 100 pmol maximum, and 250 pmol for proline.
